Design Notes
The 40 base spacer was added because lacY is found downstream of lacZ (~3.1kb) in the e. coli genome. The spacer was added to ensure ample room for regulatory elements or other genes to be added upstream of it.

The ORF and RBS came from the Escherichia coli K-12 sequence on biocyc.org. The spacer contains random bases.
